Frequently Asked Questions

DNA extraction involves lysing the cells and solubilizing DNA, which is followed by chemical or enzymatic methods to remove macromolecules, lipids, RNA, or proteins.

There are 3 basic steps involved in DNA extraction, that is, lysis, precipitation and purification.

Using ice-cold water and ice-cold alcohol will increase your yield of DNA. The cold water protects the DNA by slowing down enzymes that can break it apart. The cold alcohol helps the DNA precipitate (solidify and appear) more quickly. Make sure that you started with enough DNA.

Blood contains a number of enzyme inhibitors that can interfere with downstream DNA analysis. In addition, common anticoagulants such as heparin and EDTA can interfere with downstream assays. DNA isolation from blood requires a method to provide high-quality DNA without contaminants or enzyme inhibitors.

DNA can be isolated from any material containing it either within cells as intact molecule or outside of cells as fragmented molecule. However, DNA samples from humans and many other animals are often extracted from blood or skin cells. The easiest way to collect skin cells from humans is to brush the inside of the cheek with a cotton swab. This is called a buccal swab.
